Overview
The T-cell receptor (TCR) is a heterodimeric, membrane-bound protein complex on the surface of T lymphocytes that recognizes antigenic peptides presented by major histocompatibility complex (MHC) molecules, providing the specificity that drives adaptive cellular immunity. Most T cells express an alpha-beta TCR, while a minority bear gamma-delta chains; each receptor is generated through somatic V(D)J recombination, producing the immense clonal diversity needed to recognize a vast array of antigens. The TCR itself lacks intrinsic signaling capacity and associates with the invariant CD3 complex, which transduces activation signals upon peptide-MHC engagement, with co-receptors CD4 and CD8 stabilizing the interaction and helping distinguish self from non-self. TCR signaling governs T-cell development, thymic selection, activation, proliferation, and differentiation into effector and memory subsets, and its specificity is central to immune surveillance against infected and malignant cells as well as to tolerance and autoimmunity. Understanding TCR structure, repertoire, and signaling underpins advances in immunology and immunotherapy, including engineered receptors and adoptive cell therapies designed to redirect T cells against defined antigens. Characterization of the receptor's recombination, affinity, and downstream pathways continues to inform vaccine design, transplantation, and the manipulation of T-cell responses in disease.
